چکیده
Embryogenesis proceeds by a highly regulated series of events. In animals, maternal factors that accumulate in the egg cytoplasm control cell cycle progression at initial stage cleavage. However, regulation is switched to system governed activated nuclear genome specific development, referred as maternal-to-zygotic transition (MZT). Detailed molecular analyses have been performed on and zygotic genes MZT mammals, fishes chicken; however, underlying mechanisms remain unclear quail. present study, we demonstrated occurred blastoderm V Japanese quail using novel gene targeting technology which CRISPR/Cas9 intracytoplasmic sperm injection (ICSI) systems were combined. At V, found retinoblastoma 1 (RB1) protein expression was down-regulated, whereas cyclin D1 (CCND1) initiated. When microinjection sgRNA containing CCND1-targeted sequencing Cas9 mRNA administered pronuclear stage, development stopped down-regulation RB1 did not occur. This result indicates most notable difference from mammals CCND-knockout embryos are capable developing beyond MZT. We also showed CCND1 induced phosphorylation serine/threonine residues protein, resulted degradation this protein. These results suggest one key for MZT, elimination may contribute after during Our technology, combined ICSI, has potential become powerful tool avian-targeted mutagenesis.

Maternal Argonaute 2 is essential for early mouse development at the maternal-zygotic transition.
Activation of zygotic gene expression in the two-cell mouse embryo is associated with destruction of maternally inherited transcripts, an important process for embryogenesis about which little is understood. We asked whether the Argonaute (Ago)/RNA-induced silencing complex, providing the mRNA "slicer" activity in gene silencing, might contribute to this process. Here we show that Ago2, 3, and ...
متن کاملCyclin D1 Expression in Patients with Laryngeal Squamous Cell Carcinoma
Background & Objective: Laryngeal squamous cell carcinoma (LSCC) is considered to be one of the most common cancers of the head and neck, accounting for roughly 90% of all malignant tumors of the larynx. To have a timely diagnosis for a better and practical therapy, molecular markers have to be investigated. The aim of this study was to determine the expression of Cyclin D1 (CD...
متن کاملCyclin D1 promotes mitogen-independent cell cycle progression in hepatocytes.
Cyclin D1 is widely believed to regulate progression through G1 phase of the cell cycle, and previous studies have shown that this protein is induced during hepatocyte proliferation in culture and in vivo. In this study, the role of cyclin D1 in the cell cycle of primary rat hepatocytes was further examined. Following epidermal growth factor stimulation, cyclin D1 was upregulated at time points...
متن کاملProgression in Human Breast Cancer Cells Cyclin D1 Is Necessary for Tamoxifen-Induced Cell Cycle
Despite the success of tamoxifen in treating hormoneresponsive breast cancer, its use is limited by the development of resistance to the drug. Understanding the pathways involved in the growth of tamoxifen-resistant cells may lead to new ways to treat tamoxifen-resistant breast cancer. Here, we investigate the role of cyclin D1, a mediator of estrogendependent proliferation, in growth of tamoxi...
متن کاملAn essential role for the RNA-binding protein Smaug during the Drosophila maternal-to-zygotic transition.
Genetic control of embryogenesis switches from the maternal to the zygotic genome during the maternal-to-zygotic transition (MZT), when maternal mRNAs are destroyed, high-level zygotic transcription is initiated, the replication checkpoint is activated and the cell cycle slows. The midblastula transition (MBT) is the first morphological event that requires zygotic gene expression. The Drosophil...
